Univestin is composed of plant extracts standardized for specific bioflavonoids from two botanicals with a long history of safe human consumption, Scutellaria baicalensis and Acacia catechu (Senegalia catechu). The Univestin plant extracts are standardized for two distinct types of bioflavonoids that Unigen discovered to inhibit the pro-inflammatory enzymes COX and LO and to also neutralize inflammatory reactive oxygen species (ROS). Univestin was shown to be safe and efficacious in extensive preclinical animal testing and human clinical trials. Over 1230 medicinal plants from Unigen’s PhytoLogixTM library were screened to identify natural substances with COX and LO dual inhibitory activity. Following confirmatory assays with freshly prepared materials and dose response titrations, the best of the “hits” from the screen were selected for assay-guided HPLC fractionations to isolate pure active compounds. The activity of purified compounds was verified with enzyme inhibition assays and cellular assays for inhibition of Arachidonic Acid (AA) metabolite production. The identities of the active compounds were then established by MS, NMR and natural product database references. Based on these in vitro assays, efficacious extracts were then tested for inhibition of inflammation in vivo using both air pouch and ear-swelling inflammation models with extracts administered both IP and orally. The Univestin bioflavonoids, baicalin from Scutellaria baicalensis and catechins from Acacia catechu (Senegalia catechu), were identified as the active components of the most effective extracts tested in vivo. The combination of these two extracts produced a synergistic analgesic and anti-inflammatory effect in vivo with regard to both onset and duration.
